Description
Product overview
Semaglutide is a synthetic 30 amino-acid analogue of glucagon-like peptide-1 (GLP-1) marketed as Ozempic (type 2 diabetes), Wegovy (chronic weight management), and Rybelsus (oral diabetes formulation). Supplied as 5mg lyophilised per vial. The most-validated GLP-1 receptor agonist in published clinical research.
Structural engineering
Native GLP-1 with two modifications: Aib substitution at position 8 (DPP-IV resistance), Arg substitution at position 34, and a C18 fatty diacid sidechain attached via glutamate linker that binds serum albumin. The albumin binding extends plasma half-life from minutes to approximately 7 days.
Research mechanism
- GLP-1 receptor activation in pancreatic β-cells, hypothalamus, brainstem
- Glucose-dependent insulin secretion
- Delayed gastric emptying
- Central appetite suppression via hypothalamic and brainstem receptors
Trial data summary
STEP 1 trial (Wilding et al., NEJM 2021): mean body weight reduction of 14.9% at 68 weeks with weekly 2.4mg dosing. Consistent results across the STEP programme.
See our Ozempic mechanism article and semaglutide brand comparison for detailed trial data.
Research applications
- GLP-1 receptor pharmacology
- Glucose homeostasis research
- Weight regulation models
- Cardiovascular outcome research
Reconstitution and storage
Reconstitute with bacteriostatic water at 1–2 mL for working concentrations of 2.5–5 mg/mL. Refrigerate (2–8°C), stable approximately 4 weeks after reconstitution. Store lyophilised vial at –20°C until reconstitution.
Quality and analytics
HPLC purity ≥98%. Batch-specific Certificate of Analysis with mass spectrometry confirmation available on request.
For laboratory research use only. Not for human or veterinary use.
