Melanotan II (MT-II) is a synthetic cyclic heptapeptide analogue of α-melanocyte stimulating hormone (α-MSH). Originally developed at the University of Arizona as a photoprotective research candidate. Supplied as 10mg lyophilised per vial.
Sequence and structure
Ac-Nle-cyclo(Asp-His-D-Phe-Arg-Trp-Lys)-NH₂. The cyclic backbone (lactam bridge Asp²-Lys⁷) confers protease resistance and locks the active receptor-binding conformation.
Research mechanism
Non-selective agonist across the melanocortin receptor family (MC1R, MC3R, MC4R, MC5R) with comparable potency at each. Activates adenylyl cyclase, elevates intracellular cAMP, and triggers downstream pathway-specific effects including:
- MC1R: melanogenesis via MITF upregulation and tyrosinase induction
- MC4R: appetite and sexual response pathway modulation
- MC3R: energy homeostasis and inflammation modulation
- MC5R: exocrine and sebaceous function

Reconstitution and storage
Reconstitute with bacteriostatic water at 1–2 mL for working concentrations of 5–10 mg/mL. The cyclic backbone provides good stability. Refrigerate (2–8°C), stable approximately 4 weeks. Protect from light to prevent tryptophan photodegradation.
